This may be caused by a number of issues, so to be sure, here are some ideas for you:
1. Turn the Tab off and then on again.
2. Go into recovery mode and wipe cache partition. You may refer to this guide: https://www.youtube.com/watch?v=Ue0vfxpU5BE
3. If the S Pen doesn't connect after steps 1 and 2, try a factory data reset (taking care to back up your data before doing so.)
Lastly, if none of these work, you may have a faulty device. Please bring both the Tab S6 and its S Pen to your nearest Samsung Service Center, or pre-book an appointment via your Samsung Members app.
