Have to tried running the diagnostic tool on the forums??

All, if not most Samsung Phones have stereo microphone. Once you use the loudspeaker, I'm sure it switches to the mic up top of your phone. (someone can rebut me on that if that is not the case.)
If they can't hear you if its on loudspeaker, then try to make sure you have tried talking in both microphones, up and bottom. If none of them works, then it is a hardware issue...
Or try and use the ear-piece. If they still can not hear you, then it is definitely a hardware problem. If they can however, it might be a random and rare bug that the microphone randomly stops working whenever on loudspeaker mode.

@EnZ143PoGi thank you for your advice

@shr17, as the members advised, if the microphone is not working when you are on speaker all the time, it is most likely a hardware issue. Make sure you are not muting yourself with the mute button when on calls.
Because the problem is most likely hardware, this issue is difficult to troubleshoot without physically inspecting your device. Please contact your local Samsung Service Center (http://www.samsung.com/ca/support/servicelocation/). Thanks!
